Spettrofotometro FTIR ad alte prestazioni Varian 670-IR

Breve descrizione della strumentazione
La spettroscopia infrarossa a trasformata di Fourier (FTIR) permette di ottenere informazioni sulla struttura delle biomolecole attraverso l’analisi del loro assorbimento nella regione spettrale del medio infrarosso. Il campione può essere esaminato sia in soluzione che in forma solida (liofili, film idrati) grazie alla possibilità di effettuare misure FTIR sia in trasmissione che in riflessione totale attenuata (ATR). Inoltre, principalmente attraverso misure di microscopia FTIR, ottenuta accoppiando il microscopio IR allo spettrometro FTIR, è possibile analizzare cellule intatte, tessuti e biofluidi per ottenere un’impronta spettroscopica del campione che fornisce informazioni sulla presenza e struttura delle principali biomolecole. Nel nostro Dipartimento è presente lo spettrometro FTIR ad alte prestazioni Varian 670-IR (Varian Australia Pty Ltd, Mulgrave VIC, Australia) con allineamento dinamico e rivelatore ad alta sensibilità (MCT). E’ dotato di cella porta-campione per misure in trasmissione e sistema ATR (in diamante a singola e a nove riflessioni) con controllo della temperatura. Lo spettrometro è accoppiato al microscopio IR Varian 610-IR. La spettroscopia FTIR permette di ottenere informazioni complementari a quelle che si possono ottenere con altri approcci strumentali disponibili presso il nostro Dipartimento. Ad esempio, nel caso delle proprietà conformazionali e stabilità di proteine e peptidi è possibile eseguire caratterizzazioni di spettroscopia FTIR, di dicroismo circolare e fluorescenza intrinseca ed estrinseca.
